反义寡核苷酸抑制端粒酶活性对喉癌细胞癌基因表达的影响 被引量:5

摘要 目的 探讨针对端粒酶的反义寡核苷酸对喉癌细胞端粒酶活性的抑制及对P53和PCNA基因表达的影响。方法 用PCR ELISA法检测喉癌细胞株Hep 2细胞内端粒酶的活性。用免疫组化的方法检测喉癌细胞P53和PCNA基因的表达。结果 该反义寡核苷酸可抑制喉癌细胞端粒酶活性 ,此作用有序列特异性和浓度依赖性。 40 μM反义寡核苷酸抑制了喉癌细胞PC NA的表达 ,而对P53表达无明显影响。结论 靶向于端粒酶的反义寡核苷酸能抑制喉癌细胞Hep 2PCNA基因的表达。 Objective To study the inhibition of telomerase activity and the influence of expression of carcinoma gene to laryngeal cacinoma cells incubated with AS ODN targeting against telomerase in vitro.Methods Telomerase activity of laryngeal carcinoma cells Hep 2 was analysised by PCR ELISA technique.Expression of P53 and PCNA was detected by immunohistochemical method.Results The AS ODN inhibited telomerase activity of laryngeal carcinoma cells.This procedure is sequential specific and density dependence. The 40μM AS ODN inhibited the expression of PCNA, while had no significant effect on expression of P53 in laryngeal cacinoma cells.Conclusion The AS ODN targeting against telomerase inhibited the expression of PCNA gene in laryngeal carcinoma cells.
