摘要
目的 探讨TGF - β及HSP70在人重组白细胞介素 - 1(rhIL - 1β)介导颞颌关节炎症损伤中的作用。方法 SD大鼠颞下颌关节腔内反复多次注射rhIL - 1β,建立颞颌关节炎症损伤模型 ,利用免疫组织化学检测髁状突软骨炎症损伤过程中TGF - β及HSP70的表达变化。 结果 注射后 1- 3d ,实验侧髁状突软骨全层均可见TGF - β阳性软骨细胞 ,以增殖层阳性表达最明显 ,7d除髁状突软骨增殖层表达仍较强 ,前肥厚层TGF - β表达开始减弱 ,30d髁状突软骨全层TGF - β表达与对照侧相近。HSP70的表达与TGF - β具有相似的特点 ,注射后 1-3d ,实验侧髁状突软骨浅层HSP70的表达明显增强 ,7d后表达逐渐减弱 ,15d后HSP70表达与对照侧相近。结论在rhIL - 1β介导的髁状突软骨关节炎症损伤中 ,早期TGF - β与HSP70的表达增强 ,提示TGF - β及HSP70可能参与了软骨炎症损伤的修复过程。
Objective To evaluate the role of TGF-β and HSP70 in the inflammatory lesion of temporomandibular joint.Methods The model of inflammatory lesion of TMJ in SD rats was established by intraarticular injection with recombinant human interleukin-1β.The expressions of TGF-β and HSP70 in cartilage were examined by immunohistochemical staining.Results rhIL-1β injection resulted in inflammatory reaction in TMJ,and destruction of condylar cartilage was found;the expressions of TGF-β and HSP70 proteins increased in condylar cartilage during 1~7days of injection and decreased gradually after rhIL-1β injection 15 days later.Conclusion The results indicate that the functions of TGF-β and HSP70 are activated in TMJ inflammatory lesion,which may play an important role in the destructive process of OA.
