摘要
目的 观察组合型生物人工肝的临床应用效果。方法 应用两步胶原酶消化法分离猪肝细胞 ,成功构建生物反应器和组合型生物人工肝 ,并将之应用于 2例中晚期重型肝炎患者 ,共治疗 3次。结果 应用构建的生物型生物人工肝治疗时患者生命体征平稳 ,血清谷丙转氨酶 (ALT)、谷草转氨酶 (AST)、总胆红素 (TBI)、凝血酶原时间 (PT)及凝血酶原活动度 (PTA)等生化指标治疗后均有一定程度改善 ,治疗后 2天 ,ALT、AST、PT及PTA仍持续好转。结论 组合型生物人工肝脏使用安全 。
Objective To study the effects of hybrid bioartificial liver in middle and end stage of severe hepatitis.Methods Liver cells were isolated from the parenchyma of the pig liver using selgen's enzymatic collagenase perfusion methods.Bioreactor was constructed by placing the isolated liver cells on the extra capillary side of the fibres in a hollow fibre device and forming a recycling liver cell loop.Hybrid bioartificial liver was developed by connecting the bioreactor with the non bioartificial devices.Two cases of middle or end stage of severe hepatitis were treated with this hybrid bioartificial liver 3 times.Results The vital signs of the two patients remained stable during the treating periods.After the support of hybrid bioartificial liver,the transaminase,total bilirubin index,prothrombin time and prothrombin time activity were all improved to some extent.Two days later,the transaminase?prothrombin time and prothrombin time activity were further improved.Conclusion The application of our hybrid bioartificial liver is safe and effective.
