摘要
内毒素即脂多糖介导的细胞信号转导在激活靶细胞产生一系列应答反应、导致机体损伤并出现病理变化中起着关键作用。LP S信号转导涉及到多种血清蛋白和受体,如LBP、sCD14、mCD14、以及CD11/CD18。LBP/CD1 4系 统在LPS信号转导中起着重要作用,但并非LPS信号转导所必需的。LPS与受体结合后,细 胞 内信号转导涉及到酪氨酸激酶与MAPK超家族,通过多个MAPK通路和保守的三级激酶级联反 应 (MAPKKK-MAPKK-MAPK)将LPS信号转导至胞内,引起细胞应答。同时,p38 MAPK对LPS 介导 的细胞反应有重要调节作用。这些均表明,LPS信号转导可能是一个多受体、多通路、可调 控的复杂过程。
The signal transmembrane transduction induced by bacteri al endotoxin(ET ), lipopolysaccharide(LPS),plays a key role in the process that LPS activates ta rget cells to produce a series of response leading to body's injury and patholog ical changes.There are involved in several kinds of plasma proteins and receptor s such as LPS-binding protein(LBP),soluble CD14(sCD14),membrane CD14(mCD14)an d CD11/CD18 in the signal transduction of LPS.LBP/CD14 system is very important b ut unnecessary for LPS signal transduction.After LPS binds with receptor,the LPS signal transduction in the cells involves tyrosine kinase and mitogen-activate d protein kinase(MAPK) super family,the signal is transduced into cell resulting in cellular response by many MAPK pathways and conservative kinase cascade react ion(MAPKKK-MAPKK-MAPK).Meanwhile,p38MAPK has an important effect on the cellul ar response induced by LPS.All of these indicate that the signal transduction of L PS is probably regulatory complicated course with multi-receptor and multi-pat hway.
