摘要
目的 研究实验性脊髓损伤后低氧诱导因子 (HIF- 1α)的时序性变化及与创伤性凋亡的关系 ,探讨其在脊髓损伤中的意义。方法 以大鼠静压型脊髓损伤模型为研究对象 ,运用免疫组化方法进行 HIF- 1α染色 ,流式细胞术测定凋亡率及 bcl- 2的表达。结果 从脊髓损伤后 1天开始 ,HIF- 1α表达明显增加 ,3~ 7天达到高峰 ,14天时 HIF- 1α阳性染色率下降显著。 HIF- 1α的时序性变化与凋亡率和 bcl- 2含量的变动趋势密切相关。结论 HIF- 1α可能参与了脊髓损伤后缺血缺氧的继发损害过程 ,它与创伤后凋亡之间有着密切的关系 。
Objective To investigate the sequential changes of hypoxia inducible factor 1α(HIF 1α) in experimental spinal cord(SCI) injury in rats and analyze its potential effects on SCI. Methods Static compression model of SCI was employed in this study. Expressions of HIF 1α were measured with immunohistochemical staining. Flow cytometry was used to determine the apoptotic ratio and bcl 2 expressions. Results HIF 1α began to increase one day after injury, and reached peak at 3 7 days. Two weeks later, it declined significantly. The sequential changes of HIF 1α coincided well with the alternations of apoptotic ratio and bcl 2 content. Conclusion HIF 1α may participate in the secondary ischemic and hypoxic procedures after spinal cord injury and mediate the traumatic apoptosis. Further understanding of HIF 1α may provide new therapeutic regimens for SCI.
