摘要
目的 了解丹参酮 A(Tan A)对 K5 6 2细胞株的生长影响及红系诱导分化作用 ,探讨其可能的作用机制。方法 细胞培养、形态观察和流式细胞术检测等。结果 Tan A对 K5 6 2细胞有生长抑制作用 ,而适当浓度的 Tan A可诱导 K5 6 2细胞向红系分化。用 0 .5 μg/ m l的 Tan A处理后 ,K5 6 2细胞的凋亡细胞增加 ,G0 / G1 期细胞堆积 ,c- m yc、bcl- XL 和 H- ras的表达下降 ,p5 3和 Rb的表达增加。结论 Tan A对 K5 6 2细胞有生长抑制及诱导红系分化的作用 。
Objective To investigate the effect of Tanshinone ⅡA on K562 cell line and its possible mechanism. Methods Cell culture, cytomorphology and cytometrics were used. Results The proliferative inhibition of K562 cells induced by Tanshinone ⅡA was dose dependent, and the degrees of erythroid differentiation induced by 30 nmol/L ACM and 0.5μg/ml Tanshinone ⅡA were not statistically different ( P >0 05). Conclusion\ Tanshinone ⅡA has an inhibitive effect on the proliferation of K562 cells and an inductive effect on the differentiation of erythrocyte series. The mechanisms may be related to the changes of cell cycle and gene expressions.
