Study on the induction of transplantation tolerance to the allogeneic skin graft in mice by anti-MHC-Ⅰ monoclonal antibody

摘要 目的 通过静脉注射异基因脾细胞和抗H 2 b 单抗加腹腔注射环磷酰胺诱导成年B6小鼠对异基因皮肤移植物产生免疫耐受。方法 经C5 7BL/ 6 (H 2 b,B6 )小鼠尾静脉注射BALB/c小鼠 (H 2 d)脾细胞 ,2d后腹腔注射环磷酰胺 (CP) ,随后 2次尾静脉注射抗小鼠的H 2 b 单克隆抗体 ,然后进行皮肤移植 ,并于耐受 30d对受体B6小鼠作混合淋巴细胞反应 (mixedlymphocytereaction ,MLR)、迟发型超敏反应 (delayedtypehypersensitivity,DTH)等耐受状态检测。结果 BALB/c小鼠的皮肤移植物在耐受B6小鼠中存活期特异延长。MLR和DTH检验证明 :B6小鼠对BALB/c小鼠的脾细胞产生特异性耐受 ,对无关第 3者KM小鼠的脾细胞仍表现出强烈的免疫反应。结论 抗MHC Ⅰ类分子单克隆抗体可明显促进“细胞 +CP”诱导的异基因皮肤移植耐受 ; Objective To study the effect and mechanism of anti MHC Ⅰ monoclonal antibody on tolerance induced by using the “cells followed by CP' system. Methods Spleen cells of BALB/c mice (H 2 d) were injected to C57BL/6 mice (H 2b, B6) via the tail vein. Two days later an intraperitoneal injection of cyclophosphamide was given to the B6 mice, and following 2 consecutive anti H 2b monoclonal antibody injection via the tail vein, skin grafts of BALB/c mice were transplanted to the B6 mice. 30 days after induction of tolerance the MLR and DTH of tolerance B6 mice were examined to confirm the tolerance status. Results The survival time of allogenic skin grafts in the tolerant B6 mice was prolonged significantly. The results of MLR and DTH indicated that B6 mice were tolerant to the spleen cell of BALB/c mice, but not KM mice. Conclusions Anti H 2 b monoclonal antibody could improve the tolerance of allogenic skin graft induced by 'cells followed by CP' system. Clonal deletion is the main mechanism of the tolerance. [
