摘要
目的 从基因转录水平确定人眼睫状体无色素上皮细胞是否具有合成心钠素(artial natriuretic peptide,ANP)的生物学特性。方法 体外培养新鲜成人尸眼和流产胎儿眼睫状体无色素上皮细胞 ,从中抽取总 RNA ,应用逆转录 -聚合酶链反应 (reverse tran-scription- polymerase chain reaction,RT- PCR)技术 ,对睫状体无色素上皮细胞的 m RNA进行检测。结果 体外培养的人眼睫状体无色素上皮细胞中存在有 ANP m RNA的表达。结论 人眼睫状体无色素上皮细胞有合成 ANP的功能 ,它可能在眼压的调节过程中起着重要作用。
Objective To determine whether human nonpigmented ciliary epithelial cells can physiologically synthesize artial natriuretic peptide (ANP) on the transcriptional level. Methods Human nonpigmented ciliary epithelial cells were isolated and cultured in vitro. Total RNA was extracted from those cells when primary culture became confluent. ANP message RNA(mRNA) was identified by reverse transcription-polymerase chain reaction (RT-PCR). Results ANP mRNA existed in human nonpigmented ciliary epithelial cells. Conclusion Human nonpigmented ciliary epithelial cells can synthesize ANP ,which may play an important role in the regulation of intraocular pressure.
