摘要
目的 观察人胎儿不同发育阶段晶状体上皮细胞凋亡 (APO) ,以及增殖细胞核抗原 (PCNA )的表达特征 ,并探讨细胞增殖与凋亡在胎儿晶状体发育中的关系。方法 用原位末端标记法 ,检测了 10~ 32周人胎儿晶状体上皮细胞凋亡阳性表达率 ,用 SP免疫细胞化学技术检测了同期人胎儿晶状体上皮细胞 PCNA阳性表达率。结果 PCNA和 APO在人胎儿不同胎龄晶状体上皮细胞均有不同程度的表达 ,阳性细胞核为桔黄色或棕黄色。在胎儿晶状体发育的早期 (10~ 12周 )细胞增殖阳性率明显高于凋亡 ,晶状体上皮细胞以增殖为主 ;在胎儿晶状体发育的中期 (16~ 2 8周 )细胞凋亡阳性率明显高于增殖 ,晶状体上皮细胞凋亡占优势 ;在晶状体发育的晚期 (32周 ) ,细胞增殖与凋亡活动均较弱。结论 细胞凋亡与增殖行为在胎儿晶状体发育的整个过程中普遍存在 ,协同参与了调控胎儿晶状体正常发育过程。
Objective To observe apoptosis (APO) and the expressing features of proliferating cell nuclear antigen (PCNA) in lens epithelial cells (LECs) in different stages of human fetal development, in order to approach the relationship between proliferation and APO in the course of lens development. Methods The positive rate of APO and PCNA in LECs of human fetus were examined from 10th to 32nd week by using TdT~mediated dUTP nick end labeling(TUNEL) and SP immunocytochemistry respectively. Results Different degree expressions of PCNA and APO were found in LECs at different fetal ages. Positive nucleus was orange or brown. In the early stage of lens development (from 10th to 12th week), the positive rate of PCNA was higher than APO, and proliferation was the major expression. In the medium term of lens development (from 16th to 28th week), the positive rate of APO was higher than PCNA, and APO was the major expression. In the later period of lens development (32nd week), the activities of proliferation and APO were both weak. Conclusion Proliferation and APO exist universally in the course of lens development, and supervise and control the regular development of lens co-operatively.
