摘要
目的观察反应停能否抑制慢性肝炎外周血单核细胞 ( peripheralbloodmononuclearcells,PBM Cs)肿瘤坏死因子α(TNFα)的产生 ,明确反应停对慢性肝炎的治疗作用。方法用淋巴细胞分离液从 1 3例慢性肝炎患者的外周血中分离PBMCs进行体外培养 ,用酶联免疫吸附法、免疫组化法和半定量斑点杂交法 ,观察反应停对PBMCs肿瘤坏死因子和其它细胞因子的产生和基因表达的影响。结果反应停可抑制PBMCsTNFα和其它细胞因子的产生以及TNFαmRNA的表达 (P <0 .0 5 )。结论反应停对慢性肝炎的治疗有重大的意义。
Objective To study whether thalidomide (Thal) has the same effects on tumor-necrosis factor α (TNFα) in chroinc hepatitis as it does in many other inflammatory diseases, can reduce the production of TNFα by mononuclear cells.Method Peripheral blood mononuclear cells (PBMCs) were isolated from 13 patients with chronic hepatitis by density gradient centrifugation,then cultured in vitro, or stimulated by lipopolysaccharide (LPS, 1μg/ml), the effects of thal (4μg/ml) on the syntheses and secretions of TNFα and other cytokines were studied with enzyme-linked immunosorbent assay and immunohistochemical examintion. The expression level of TNFα mRNA was evaluated with Dot-Blot hybridization combined with semiquantative assay.Results Thal could reduce the syntheses and secretions of TNFα (P<0.05) and other cytokines,including IL-1β, IL-6 and IL-8 induced by LPS(P<0.05);suppressed the expression of TNFα mRNA (P<0.05).Conclusions Thal or its analogues may have potential as a new therapeutics for the treatment of chronic hepatitis.
