内皮素、胰高血糖素、血管紧张素Ⅱ、醛固酮与实验性大鼠肝纤维化 被引量:3

The relationship of endothelin, glucagon, angiotensinⅡ, aldosterone and experimental rat liver fibrosis

摘要 目的 观察内皮素 (ET)、胰高血糖素 (GLUCA)、血管紧张素Ⅱ (ATⅡ )和醛固酮 (ALD)与大鼠肝纤维化形成的关系。方法 建立复合病因致大鼠肝纤维化的模型 ,分别于 2、4、6、8周末取一组大鼠 ,测定其血液及肝组织匀浆中ET、GLUCA、ATⅡ和ALD含量 ,并观察了大鼠肝脏组织学变化规律及电镜下Ito细胞数量、形态的改变 ;分析以上 4种激素与肝脏组织学改变的相关性。结果 在造模的过程中 ,大鼠血液及肝组织匀浆中ET、GLUCA、ATⅡ和ALD均不断升高 ,肝纤维化程度不断加重 ,2、4、6、8周时大鼠肝脏组织学评分分别为 :0 .6 3± 0 .5 2 ,1.75± 0 .71,2 .6 3± 0 .92和 3 .2 2± 0 .83 ;血液中ET、GLUCA、ATⅡ和ALD均与肝组织纤维化程度呈正相关 ,相关系数分别为 0 .36 0 (P <0 .0 5 )、0 .6 97(P <0 .0 0 1)、0 .72 0 (P <0 .0 0 1)和 0 .778(P <0 .0 0 1) ,肝组织匀浆中ET、GLUCA、ATⅡ和ALD也均与肝组织纤维化程度正相关 ,相关系数分别为 0 .6 82 (P <0 .0 0 1)、0 .479(P <0 .0 1)、0 .6 2 4(P <0 .0 0 1)和 0 .5 5 5 (P <0 .0 0 1)。电镜下可见模型组肝脏窦周Ito细胞被激活 ,数量显著增多 ,形态亦有明显改变。结论 ET、GLUCA。 Objective To investigate the relationship of endothelin(ET), glucagon(GLUCA), angiotensinⅡ(ATⅡ), aldosterone(ALD) and liver fibrosis of the rat model. Methods 50 male SD rats were divided into control (8 rats) and model groups (42 rats). The concentrations of ET, GLUCA, ATⅡ, ALD in the rats blood and liver tissues were measured at the end of 2nd, 4th, 6th, 8th week. Histological changes of the liver was also evaluated. The changes in number and morphological features of the Ito cells were observed under electronmicroscope. Results The concentrations of ET, GLUCA, ATⅡ, ALD in the rats blood and liver tissues of model groups increased gradually. The histological scoring of liver fibrosis at the end of 2nd, 4th, 6th, 8th week were 0.63±0.52, 1.75±0.71, 2.63±0.92, 3.22±0.83, respectively. The correlative coefficients of blood and liver tissues, the concentrations of ET, GLUCA, ATⅡ, ALD and histological scoring of liver fibrosis were 0.360( P <0.05), 0.697( P < 0.001 ), 0.720( P <0.001), 0.778( P <0.001), 0.682( P <0.001), 0.479( P <0.01), 0.624( P <0.001), 0.555( P < 0.001 ), respectively. Conclusion ET, GLUCA, ATⅡ, ALD all correlate with the liver fibrosis of rat model.
