脂质体介导的c-myc反义硫代寡核苷酸诱导胃癌SGC-7901细胞凋亡

摘要 目的 探讨c myc反义硫代寡核苷酸 (antisensephosphorothioateoligodeoxynucleotide ,APSODN)对胃癌SGC 790 1细胞凋亡的影响。方法 人工合成c mycAPSODN ,通过脂质体转染法处理人胃癌细胞系SGC 790 1后 ,于不同时间收集细胞 ,采用Northernblot、Westernblot检测c myc基因mRNA及蛋白表达 ,采用TUNEL方法检测凋亡指数 ,以流式细胞仪检测细胞周期。结果 TUNEL显示c mycAPSODN可诱导胃癌SGC 790 1细胞凋亡 ,并与APSODN的浓度和处理时间有关。流式细胞仪检测显示细胞阻滞于G0 /G1期。结论 c mycAP SODN可诱导胃癌细胞凋亡。 Purpose To study the effects of c-myc antisense phosphorothioate oligodeoxynucleotide (APSODN) on apoptosis of gastric cancer SGC-7901 cell. Methods After gastric cancer SGC-7901 cell were treated by c-myc APSODN, we studied the expression of c-myc gene using Northern blot and Western blot, apoptosis using terminal uridine deoxynucleotidyl nick end labeling(TUNEL), cell cycle using flow cytometer. Results c-myc APSODN could induce apoptosis of gastric cancer SGC-7901 cell, simultaneously, G0/G1 arrest, and the effect of induction was related to trearted time and density. Conclusions c-myc APSODN can induce apoptosis of gactric cancer cell.
