鞘内注射孤啡肽对正常及创伤大鼠NK细胞活性的影响

Effects of Intrathecal Injection of Orphanin FQ on the NK Cell Activity of Normal and Traumatic Rats

目的 观察鞘内注射孤啡肽 (orphaninFQ ,OFQ)对正常及手术创伤大鼠NK细胞活性的影响。 方法 用3H TdR释放法测定大鼠脾脏自然杀伤细胞 (NK细胞 )活性。结果 鞘内分别注射 0 .1、1μgOFQ后 ,正常大鼠NK细胞活性与生理盐水组 (NS)相比无明显差异 ;5、10 μgOFQ则明显抑制NK细胞活性 (P <0 .0 1) ,提示脊髓OFQ对正常大鼠免疫功能具有抑制作用。手术创伤后大鼠NK细胞活性明显降低 ,鞘内分别注射OFQ 0 .1、1、5、10 μg后NK细胞活性与单纯创伤组无明显差异 ,注射 2 0 μgOFQ后 ,NK细胞活性则比创伤组显著降低 ,提示鞘内注射OFQ不能翻转创伤介导的免疫抑制 ,大剂量的OFQ反而加剧免疫抑制。

Purpose: To investigate the influence of intrathecal(ith) injection of orphanin FQ(OFQ) on the immune function. Methods: NK cell activity of normal and traumatic rats were measured by 3H-TdR releasing method. Results: The results showed that ith injection of 0.1 or 1 μg OFQ had no effect on the NK cell activity, but ith 5 or 10 μg OFQ could significantly inhibit the NK cell activity in normal rats(P< 0.01). When ith 0.1,1,5 or 10 μg OFQ in the traumatic rats, the NK cell activity had no change compared with the trauma group, but ith 20 μg OFQ further reduced the immunosuppression produced by surgical trauma(P<0.01). Conclusions: These results showed that ith injection of OFQ only produced an inhibitory effect on the NK cell activity of normal and traumatic rats.
