摘要
目的 考察人在头低位卧床 (- 6°)模拟失重状态下 ,口服扑热息痛后药代动力学参数的变化 ,以指导载人航天中的合理用药。方法 用人在头低位卧床 (- 6°)模拟航天中的失重状态 ,口服扑热息痛后测定唾液中的药物浓度 ,从而推算血浆中的药物浓度 ,根据血药浓度来计算Cmax、Tmax、AUC(0~ 12h) 。结果 人在头低位卧床 (- 6°)模拟失重状态下 ,Tmax显著延长 ,而Cmax和AUC(0 -12h) 变化不明显 ,直到第 19d才显著降低。结论 头低位卧床 (- 6°)
Aim To observe the effect of -6°head-down bed-rest on the pharmacokinetic parameters of paracetamol. Methods To simulate weightlessness with -6°head-down bed-rest, the concentration of paracetamol in saliva samples were measured in order to establish the drug concentration in blood. The pharmacokinetic parameters of paracetamol, C max , T max and AUC (0-12h) ,were calculated by the drug concentration in blood. Results The Tmax was significantly extended during bed-rest; The C max and AUC (0-12h) were markedly decreased until 19 days later. Conclusions -6°head-down bed-rest has significant effect on the pharmacokinetic parameters of paracetamol.
