摘要
目的 建立高效液相色谱法 (HPLC)测定血清中艾司洛尔浓度的方法。方法 以甲醇 -冰醋酸 - 0 .2 %醋酸钠 (6 0∶1∶40 )为流动相 ,流速 1.0ml/min ,检测波长为 2 80nm。结果 在 10~ 6 0 0 μg·ml-1范围内样品色谱峰高与血清药物浓度呈良好的线性关系 ,r=0 .9998。日内及日间 RSD 分别测得为 3 .0 5 % ,4.43%。平均加样回收率为 10 0 .45 % ,血清中药物最低检测浓度为 13 .3μg·L-1。结论 本方法适于艾司洛尔的血药浓度监测。
Aim To establish a HPLC method for the determination of esmolol in Wistar rat serum. Methods The UV detector was set at 280 nm. The mobile phase consisted of methanol,acetic acid and 0.2%sodium acetate(60∶1∶40). Results The good linearity was from 10 to 600μg·ml -1 . The average recovery was 100.45%.RSD within day and between days were 3.05% and 4.43% respectively. The detection limit was 13.3μg·ml -1 . Conclusion This method is enough to be used for clinical therapeutic esmolol monitoring in serum.
