人脐静脉内皮细胞在培养中呈现的一种特殊光镜结构的观察分析

Observation and Analysis on a kind ofthe Structure of the Human Umbilical VeinEndothelial Cells during the Culture under the Optical Microscope

目的 :光镜下观察分析人脐静脉内皮细胞在培养过程中的一种特殊结构。方法 :经胶原酶消化 ,进行人脐静脉内皮细胞的原代培养 ,Giemsa染色制得玻片标本。结果 :在透射电镜观察及Ⅷ因子免疫组化检测证实的基础上 ,光镜下观察培养至 6天的标本 ,可见多个细胞突起汇合 ,形成“管状”结构。结论 :人脐静脉内皮细胞在培养中的这种光镜结构 ,有别于平滑肌细胞和成纤维细胞。

Objective:To observe and analyse a kind of the structure of the cultured human umbilical vein endothelial cells(HUVEC) under the optical microscope.Method:The primary culture of HUVEC digested by collagenase was performed,obtaining the examples of the glass slide of the cultured cells by Giemsa staining.Result:The confluences of the protuberance of several cells could be observed under the optical microscope on the basis of being comfirmed by the observation of the transmission electron microscope and the examination of the immunohistochemical staining of the factor Ⅷ related antigen(F Ⅷ R) when the cells were cultured about 6 days,forming the “tubular” structure.Conclusion:The cultured HUVEC was different from the smooth muscle cells and the fibroblast on the special structure under the optical microscope.
