摘要
目的:探讨地塞米松对小梁细胞生长周期的影响。方法:按常规方法培养人眼小梁细胞,然后分为二组:常规DMEM培养液及含10-7M地塞米松的DMEM培养液组,培养3d、5d、7d后,应用流式细胞仪检测细胞周期各阶段G1、S、G2期情况。结果:在正常未用地塞米松处理组,G2期细胞为2.0%;地塞米松处理 3、5、7d后,G2期细胞比例各为8.0%、16.3%、19.87%,其差异有显著性意义。结论:地塞米松可使小梁细胞生长周期中G2期的比例增加,提示这种作用可能与激素性青光眼的发病有关。眼科学报2001;17:103~105。
Purpose: To investigate the influence of dexamethesone (Dex) on cell cycle in humantrabecular cells (HTC).Methods: The trabecular specimens from human eyes were cultured in vitro. Then thecultured cells were immersed in DMEM solution containing 10 ~7M dexamethesone forthree, five and seven days. The cell cycle was detected by Flow Cycometry.Results: the Ga phase of cell cycle in HTC increased when HTC were treated by Dex.The &2 phase was 2% in control group. For three, five and seven days treated by Dex,the G2 phase were 8% , 16. 3 and 19. 4% .Conclusion: The influence of cell cycle might be induced by Dex, it may be related withthe pathogenesis of glucocorticoid induced glaucoma.
