氯胺酮对大鼠大脑皮层和丘脑突触体Na^+K^+-ATP酶活性的影响 被引量:3

EFFECTS OF KETAMINE ON Na^+K^+-ATPASE ACTIVITY OF CEREBROCORTEX AND THALUMUS IN THE RAT BRAIN SYNAPTOSOMES IN VIVO

摘要 目的 动态观察氯胺酮对大鼠大脑皮层和丘脑Na+,K+-ATP酶活性的影响。方法 SD大鼠 32只 ,随机分为对照组、麻醉组、恢复Ⅰ组和恢复Ⅱ组。对照组腹腔注射生理盐水 10ml·kg- 1,其余各组均为腹腔注射氯胺酮 10 0mg·kg- 1。对照组腹腔注射生理盐水后 10min断头 ,麻醉组、恢复Ⅰ组和恢复Ⅱ组分别在大鼠翻正反射消失后 10min、翻正反射恢复后和完全清醒后断头 ,取双侧大脑皮层和丘脑匀浆 ,离心 ,制备粗制突触体 ,用分光光度法测Na+,K+-ATP酶活性。结果 大鼠氯胺酮 10 0mg·kg- 1腹腔注射能明显降低大脑皮层和丘脑Na+,K+-ATP酶活性 ,分别较对照组降低了 32 8%和 31 4 % (P <0 0 5 ) ,而在翻正反射恢复后和动物完全清醒后Na+,K+-ATP酶活性恢复 (与对照组相比 ,P >0 0 5 )。结论 Na+。 Objective To investigate the effects of ketamine on Na +,K +-ATPase activity of cerebrocortex and thalumus in the rat brain in vivo.Method Thirty-two SD rats were divided randomly into four groups:control group,anefhesig group,recovery I group and recovery Ⅱ group.The rats in control group were administered intraperitoneally (ip) normal saline 100ml·kg -1 and decapited after 10min.The others were administered ketamine 100mg·kg -1 ip and then decapited respectively at the time of losing righting reflex,recovering righting reflex and completely aware in anesthesia group,recovery Ⅰ group and recovery Ⅱ group.The brain tissues were rapidly dessected into cerebrocortex and thalums on the saline ice and then homogenized and centrifuged.Na +,K +-ATPase activity were assessed by spectrophotometric analysis.Result After ketamine 100mg·kg -1 were administered,the Na +,K +-ATPase activity of cerebrocortex and thalumus in the rat brain were decreased by 32.8%and 31.4% as compared with control group(P<0.05).The Na +,K +-ATPase activiry of cerebrocortex and thalumus in the rat brain recovered in recovery Ⅰ group and recovery Ⅱ group(P>0.05 vs. control group).Conclusion These results indicate that Na +,K +-ATPase may play a important role in the mechanism of ketamine anaesthesia.
