原位杂交检测EGFR和c-erbB-2基因在人脑胶质瘤中的表达 被引量:4

摘要 目的 探讨表皮生长因子受体 (EGFR)和 c- erb B- 2基因表达与人脑胶质瘤发生发展的关系 .方法 采用原位杂交方法检测 5 2例人脑胶质瘤、 2株人脑胶质瘤细胞系及 8例正常人脑组织中 EGFR和 c- erb B- 2基因 m RNA的表达 .结果  EGFR m RNA阳性率在高恶性度 ( ~ 级 )胶质瘤为 79.2 % ,低恶性度 ( ~ 级 )胶质瘤为 5 0 .0 % ,二者差异显著 (P<0 .0 5 ) ;c- erb B- 2 m RNA阳性率在高恶性度胶质瘤为 83.3% ,低恶性度胶质瘤为 71.4% ,二者无显著差别 (P>0 .0 5 ) ;EGFR和 c- erb B- 2 m RNA表达均随胶质瘤恶性程度增高而增强 (P<0 .0 1) .EGFR和 c- erb B- 2 m RNA在 2株人脑胶质瘤细胞系中均有表达 ,在 8例正常人脑组织中均无表达 . EGFR和 c- erb B- 2 m RNA表达结果密切相关 (P<0 .0 5 ) .结论  EGFR和 c- erb B- 2基因在人脑胶质瘤中普遍表达 ,且随胶质瘤恶性程度增高而表达增强 ,EGFR和 c- erb B- AIM To research the role of expression of epider mal growth factor receptor (EGFR) and c erb B 2 genes in the initiation and progression of human gliomas. METHODS The expression of EGFR and c erb B 2 genes was investigated by using in situ hybridization in 52 cases of gliomas, 2 glioma cell lines, and 8 cases of normal brain tissues. RESULTS The positive rate of EGFR mRNA was 79.2% and 50.0% in high grade (grade Ⅲ~Ⅳ) gliomas and low grade (grade Ⅰ~Ⅱ) gliomas respectively, significant difference was observed between the two groups ( P <0.05). The positive rate of c erb B 2 mRNA in high grade gliomas was 83.3%, whereas 71.4% in low grade gliomas, no significant difference was observed between them ( P >0.05). The expression intensity of EGFR and c erb B 2 mRNA were all enhanced with the gliomas grade increasing ( P <0.01). The expression of EGFR and c erb B 2 mRNA were found in all of 2 glioma cell lines, whereas no were in 8 cases of normal brain tissues. Their expression was closely correlated with each other ( P <0.05). CONCLUSION EGFR and c erb B 2 genes are overexpressed in human gliomas, moreover their expression are all enhanced with the gliomas grade increasing. They may play important roles in the malignant progression of human gliomas.
