肝硬化大鼠肾脏水通道蛋白2和血管加压素V2受体的表达及黄芪对其的作用 被引量:12

Expression of renal aquaporin-2 and AVP-V2 receptor in liver cirrhosis and the role of astragalus memberanaceus

摘要 目的 观察肝硬化大鼠肾脏水通道蛋白 2(AQP2)和血管加压素(AVP)-V2受体的表达情况,及中药黄芪对其的作用。方法 肾脏皮髓质 AQP2 mRNA检测采用半定量 RT-PCR法,肾脏AQP2和AVP-V2受体蛋白检测采用免疫组织化学法。结果 肝硬化 2周和 5周大鼠肾脏皮髓质AQP2 mRNA、肾脏AQP2和 AVP-V2受体蛋白表达上调,黄芪在 2周时可纠正这些指标的升高,5周时则不能。结论 肝硬化大鼠存在AVP系统和AQP2表达的异常,黄芪在早期对其有改善作用。 Objective To investigate the changes of AVP system and AQP2 in liver cirrhosis and the role of astragalus memberanaceus( AM) Methods Immunohistochemistry and relatively quantitative RT-PCR technique were used to determine the renal protein levels of AVP-V2 receptor and AQP2. and the renal levels of AQP2 mRNA of hepato cirrhotic rats induced by common bile duct ligation for 2 and 5 weeks. Results The hepato cirrhotic rats demonstrated obviously enhanced expression of AQP2 mRNA and protein, and V2 receptor protein in kidney. AM could alleviate the over expression of these parameters in compensated cirrhosis (2 weeks), but could not in decompensated cirrhosis (5 weeks) Condusion The abnormality of AVP system and AQP2 exists in hepato cirrhosis, which can be improved by AM in early stage.
