摘要 目的观察基因重组人骨形态蛋白-2(rhBMP-2)对骨质疏松性骨折愈合的作用与影响。方法选择10月龄,雌性SD大鼠60只,随机分成3个组:生理盐水对照组(Ⅰ组),rhBMP-2治疗组(Ⅱ组),密盖息治疗组(Ⅲ组),每组20只大鼠。手术方式建立骨质疏松性骨折实验模型后,Ⅱ组于骨折处注射rhBMP-2,剂量8.5μg/kg,分别于用药后2、4、8、12周进行骨痂组织X线摄片、骨痂组织骨密度(BMD)测定和生物力学测试。结果X线检查:用药后8周,Ⅲ组骨痂面积大于Ⅱ组,P<0.05。其余各时间3组骨痂面积无显著差异。骨痂组织BMD:用药后8周,Ⅲ组较Ⅱ组高,有极显著性差异,(P<0.01),而Ⅱ组与Ⅰ组比较,有显著性差异(P<0.05)。生物力学测试:用药后12周,骨标本在扭转破坏试验中,最大扭矩、扭转刚度、破坏能量方面3组比较差异无显著性。结论单独局部应用rhBMP-2治疗骨折,有促进骨折愈合的作用,采用载体方式可提高疗效。 Objective To observe the effect and influence of recombinant human bone morphogenetic protein 2 (rhBMP 2) on osteoporotic fracture healing in the rat and to expect to provide an effective therapy for osteoporotic fracture.Method 60 female 10 month old SD rats were randomized into three groups of 20 each,(rhBMP 2) treatment group and miacalcic treatment group and saline control group, After the experimental model of osteoporotic fracture by operation was established. rhBMP 2 treatment group was given rhBMP 2 8.5 μg/kg body weight local injection once.X ray photograph an bone mineral density were measured 2,4,8,12 weeks after the treatment.Result Bone area in Miacalcic group was higher than that in rhBMP 2 groups 8 weeks after the treatment(P< 0.05)and BMD in the three groups were significant difference 8 weeks after the treatment.(P< 0.05 or P< 0.01).Conclusion Recombinant human BMP 2 may stimulated osteoporotic fracture healing in the rat. But need depend on vector.
