摘要
目的 :检测黑素瘤抗原 - 1(MAGE- 1) m RNA及其编码的肿瘤抗原在非小细胞肺癌 (NSCL C)中的表达 ,探讨以该抗原作为疫苗对 NSCL C患者进行免疫治疗的可能性。方法 :用 RT- PCR方法和流式细胞术 (FCM)对 3种人肺癌细胞系、5 6例NSCL C标本和相邻正常肺组织标本 MAGE- 1m RNA及蛋白的表达情况进行检测。结果 :MAGE- 1m RNA及蛋白在 3种人肺癌细胞系均表达 ;5 6例 NSCL C标本中 ,2 7例表达 MAGE- 1m RNA ,2 2例表达 MAGE- 1蛋白 ,而在相邻正常肺组织中均不表达。 结论 :MAGE- 1m RNA及蛋白在 NSCL C中具有较高的表达频率 ,提示该抗原有望成为对 NSCL C患者进行免疫治疗的适宜靶点。
Objective: To determine the expression of melanoma antigen-1(MAGE-1) mRNA and its encoding tumor antigen in non-small cell lung cancer (NSCLC) and to explore the possibility of using it as a vaccine for immunotherapy in NSCLC patients. Methods: The expression of MAGE-1 mRNA and protein in 3 human lung cancer cell lines and 56 NSCLC samples together with the adjacent normal lung samples was examined by RT-PCR method and flow cytometry (FCM). Results: All 3 human lung cancer cell lines expressed MAGE-1 mRNA and protein. Of the 56 NSCLC samples, 27 expressed MAGE-1 mRNA and 22 expressed MAGE-1 protein, whereas none of the adjacent normal lung samples expressed MAGE-1 mRNA or protein. Conclusion: MAGE-1 mRNA and protein are expressed at a high frequency in NSCLC samples, and this antigen may be the suitable target for immunotherapy for NSCLC patients.
