HSV-TK/GCV系统治疗鼠膀胱癌细胞T739旁观者效应的实验研究

摘要 目的 为探讨HSV TK GCV系统治疗鼠膀胱癌细胞T739的旁观者效应。方法 台盼蓝染色计数法对GCV单独作用T739 TK细胞、T739细胞或其两者按不同比例混合在不同容积等情况下的杀伤作用进行研究。结果 HSV TK GCV系统有效的杀伤T739 TK细胞 ,且这种作用有明显的旁观者效应 ,旁观者效应的强弱与T739 TK细胞所占比例以及细胞间接触程度成正相关。结论 HSV TK GCV系统不仅能有效的杀伤T739 TK细胞 ,而且对T739细胞有明显的旁观者效应。 Objective To investigate the bystander effects of herpes simplex virus thymidine kinase/ganciclovir (HSV TK/GCV) system on murine bladder carcinoma cell line T739 in vitro . Methods The killing effects of GCV on T739 TK cells alone, the mixture of T739 TK and T739 cells at different ratios, or the mixture of them at ratio of 1∶1 in different volumes were observed. Results The T739 cells transferred with HyTK gene were killed in the presence of GCV in a dose dependent manner and the effect was stable and great when GCV at 4.0 μmol/L. The HSV TK/GCV system killed the T739 TK cells greatly in manners of dose and time dependence. In the mixed culture of T739 TK and T739 cells at different ratios, GCV (4.0 μmol/L) killed not only T739 TK cells, but also neighboring cells without HyTK transfection by the bystander effect and the apoptotic rate was elevated with the increase of T739 TK proportion. When the 2 cells were mixed equally, the large volume the medium were, the more living cells left. The results indicated that bystander effect of HSV TK/GCV system had not only a positive correlation with the proportion of T739 TK, but also a negative correlation with the interspace of action. Conclusion HSV TK/GCV system can effectively kill the HSV TK transferred T739 TK cells and nearby T739 cells without the transfection by the bystander effect in vitro .
