摘要
目的 :探讨不同浓度的三氧化二砷 (As2 O3 )与HL 6 0细胞共反应不同时间后对其端粒酶活性表达的抑制作用。方法 :采用端粒重复序列扩增 (TRAP)—银染法检测HL 6 0细胞的端粒酶活性 ,并经Smartview生物电泳图像分析系统扫描分析。结果 :浓度分别为 0 .1μmol/L、1.0 μmol/L、5 .0 μmol/L的As2 O3 与HL 6 0细胞共培养 3天 ,HL 6 0细胞相对端粒酶活性由97.83%下降至 9.6 2 % ,且具有剂量依赖性和时间依赖性。结论 :As2 O3 可有效抑制HL 6
Objective:To investigate the inhibitory effects of arsenic trioxide(As\-2O\-3) on telomerase activity in HL 60 cells.Methods:The telomerase activity in HL 60 cells was detected by telomeric repeat amplification protocol(TRAP) silver staining and was analyzed by the specific software.The HL 60 cells were cultured in medium containing different concentration of As\-2O\-3 for different time.Results:Treated with As\-2O\-3(0.1~ 5.0 μmol/L ) for 3 days,the relative telomerase activity in HL 60 cells varied from 97.83% to 9.62% in a dose and time dependent manner.Conclusions:As\-2O\-3 can inhibit the telomerase activity of HL 60 cells significantly.
