摘要
目的 观察了环孢菌素A(CSA)体外对 18例再生障碍性贫血 (AA) ,17例骨髓增生异常综合征 (MDS)和 10例正常对照粒系和红系祖细胞 (CFU GM、CFU E)的作用。方法 采用RPMI16 4 0琼脂和McCoy’S5A甲基纤维素半固体平皿法。 结果 (1)AA和MDS患者的CFU GM和CFU E较正常对照明显减低 (P <0 0 0 0 1) ;(2 )CsA能明显促进AA和MDS患者CFU GM、CFU E的增殖 ,这种促增殖作用随CsA剂量增加而增加 ;(3)SAA患者CFU GM和CFU E对CsA的有效率较CAA高 ;(4)MDS RAEB和RAEB T型患者CFU GM、CFU E对CsA有效率较RA、RAS高 ;(5 )MDS和AA患者中CFU GM、CFU E值重、中度减低者对CsA有效率较零者高。结论 CsA体外可促进部分AA和MDS患者CFU GM、CFU
Objective Studying the effect of CSA in vitro on CFU GM,CFU E growth from 18 patients of aplastic anemia(AA) and 17 patients of myelodysplastic syndrome(MDS) Methods The bome marrow mononuclear cells were cultured in RPMI 1640 agar and McCoy'5A mythycellose medium, respectively Results (1)The growth of CFU GM and CFU E of patients with AA and MDS were significantly lower than those of normal controls (2) CSA significantly promote the coloney growth of CFU GM and CFU E of patients with AA and MDS The higher the dose of CSA,higher the colney growth of CFU GM and CFU E (3)The response rate of SAA patient to CSA was higher than that of CAA patient (4)The response rate of RAEB and RAEB T patient was higher than that of RA and RAS (5)The reoponse rate of patient with severe and moderate decreased growth of CFU GM and CFU E was higher than that with no growth of CFU GM and CFU E Conclusion CSA could promote the growth in vitro of CFU GM and CFU E of patients with AA and MDS
