摘要
目的 建立脂多糖 (LPS)对肝脏枯否细胞核因子 κB(nuclearfactor κB)活化的模型 ,探讨维生素E抗肝纤维化机制。方法 分离、培养大鼠肝脏枯否细胞 ;设LPS刺激组、维生素E干预组。电泳迁移率改变分析 (EMSA)法检测维生素E对肝脏枯否细胞NF κB活性的影响。结果 维生素E(终浓度 1mg/L)能抑制枯否细胞NF κB活性。结论 维生素E抑制枯否细胞NF
Objective To establish the model of determining nuclear factor κB (NF κB) which is stimulated by lipopolysaccharide (LPS) in Kupffer cells from rat and investigate the antifibrotic effect of vitamin E on NF κB activation using this model Methods Kupffer cells isolated from rat were cultured and divided into two grougs:LPS stimulated group;Vitamin E treated group The NF κB activity of nuclear protein extract from the Kupffer cells was measured by eletrophretic mobility shift assay (EMSA) Results Concentration of vitamin E in 1mg/L could inhinbit Kupffer cell NF κB activity Conclusion This study date indicates that vitamin E inhibite Kupffer cell NF κB activity is one of antifibrotic mechanisms
