摘要
目的 :观察生长抑素对人胰腺癌细胞株HS76 6T生长的作用 ;测定神经鞘磷脂 (sphingomyolin ,SM )、cAMP、细胞内Ca2 + 水平 ,探讨生长抑素的受体后信息传递作用。方法 :细胞培养 ,分别加入不同浓度的生长抑素 ,PACAP1-3 8。应用MTT法来观察细胞增殖程度 ;薄层层析法测定细胞内SM ,放免法测定细胞内cAMP含量 ;Fura - 2 /AM测定 [Ca2 + ]i。结果 :不同浓度的生长抑素 (10 -6~ 10 -12 mol/L)均能有效地抑制HS76 6T的生长。能抑制HS76 6T细胞内SM、cAMP生成和细胞内Ca2 + 水平 ,cAMP生成量和细胞内Ca2 + 水平与生长抑素浓度呈负相关。结论 :生长抑素能抑制HS76 6T细胞的生长 。
Objective: The paper wos to investigate the growth modulation of somatostatin(SS) on human pancreas carcinoma cell strain HS766T and by measuring the concentrations of sphingomylin(SM), intracellular cyclic adenosine monophosphae (cAMP) and free calcium, and then to find the pathways of postreceptor signal transduction. Methods: HS766T cells were cultured in DMEM medium and then were treated with the SS. Proliferation and viability of cell were measured by the Mosmanns method (MTT). Amount of SM in cells were determined by thin layer chromotograph. Intracellular cAMP levels were detected by radioimmunoassay . Intracellular Ca 2+ level was measured by using a F 2000 Flurorescerce spectrophotometer. Results: SS inhibited the proliferation of HS766T in a concentration dependent manner (P< 0.01 ,r=-0.921). Production of intracellular SM,intracellular cAMP and Ca 2+ in HS766T cells were inhibited by SS. The amount of production was negatively correlation with concentration of SS (P<0.01). Conclusion: The proliferation of HS766T were inhibited by SS. The effect of SS on human pancreatic cancer cells might be mediated by specific receptor.
