摘要
目的:探讨中药苏木提取物诱导人类慢性髓性白血病K562细胞株的凋亡作用。方法:采用MTT法检测苏木提取物对K562细胞增殖抑制作用;荧光显微镜观察细胞形态变化;库尔特全自动颗粒粒度分析药物引起K562细胞体积大小的分布变化;琼脂糖凝胶电泳测定DNAladder及流式细胞术检测细胞周期变化。结果:25μg/ml苏木浸膏能明显诱导K562细胞凋亡,产生凋亡细胞所具有的典型形态学及生化学特征,同时对K562细胞的增殖抑制和诱导凋亡作用呈一定的浓度依赖性。结论:中药苏木提取物能诱导细胞凋亡,抑制癌细胞增殖呈一定的浓度依赖关系。
Objective: To investigate whether Caesalpinia sappan L.extract compound can induce apoptosis in human leukemia cell line K562.Methods: Cell proliferation was determined by MTT assay.Morphological changes of apoptosis were observed with invert fluorescence microscope. The sizes of cell volume were assessed by Coulter Multisizer Ⅱ analytical instrument. DNA ladder and cell cycle were examined by DNA agarose gel electrophoresis and PI fluorescence flow cytometry (FCM), respectively. Results: MTT assay showed that Caesalpinia sappan L.extract with concentration from 3.12 to 25 μg/ml could inhibit cell proliferation. The inhibitory rate depended on Caesalpinia sappan L.concentration. There were obvious changes of morphology, biochemistry, and sizes of the cells after 24 h following treatment of Caesalpinia sappan L 25 μg/ml. Sub G1 peak (58.6%) and G0 G1+G2 M arrest were also determined by FCM. Conclusion: Caesalpinia sappan L.extract can induce K562 cell apoptosis and obviously inhibit cell proliferation in a dose dependent manner.
