摘要
目的 观察瘦素对大鼠肝细胞蛋白激酶 C(PKC)蛋白表达的影响。方法 分离、培养新生Sprague- Dawley大鼠肝细胞 ,与瘦素 (5 nmol/ L)孵育 12 ,2 4,36 h,提取蛋白后用 Western印迹法检测各时间点蛋白激酶 C的蛋白表达水平。结果 大鼠肝细胞 c PKCα的蛋白水平在瘦素孵育 12 h及 2 4h后同对照组相比均未发生显著变化 (P>0 .0 5 ) ;但在孵育 36 h后同对照组相比显著升高 (P<0 .0 5 )。经瘦素孵育 2 4及 36 h后 ,大鼠肝细胞 c PKCα的蛋白水平同孵育 12 h时相比亦显著升高 (P<0 .0 5 )。结论 瘦素可上调大鼠肝细胞蛋白激酶 C的蛋白表达 ,这种促进作用呈时间依赖性。
Objective To investigate the effect of leptin on the protein expression of cPKCα in rat hepatic cells. Methods After rat hepatic cells were incubated with leptin(5 nmol/L) for 12,24 and 36 hours, Western bolt was used to assess the protein level of cPKCα. Results The protein contents of cPKCα in rat hepatic cells were similar to that of control after incubated with leptin for 12 and 24 hours(P>0.05); but after incubated with leptin for 36 hours, the protein contents of cPKCα were significantly increased compared with control(P<0.05). After incubated with leptin for 24 and 36 hours, the protein contents of cPKCα were significantly increased compared with that for 12 hours(P<0.05). Conclusions The results indicated that leptin can upregulate the cPKCα protein expression in hepatic cells of rat.
