摘要
目的 探索成骨性肿瘤组织中微血管的形态及数量与瘤细胞分化、增殖的关系。方法 采用免疫组织化学染色对成骨性肿瘤中CD34和Ⅳ型胶原蛋白阳性血管计数及形态进行分析。结果 骨肉瘤以血窦型及芽状细索型血管为主 ,CD34阳性血管数多于Ⅳ型胶原阳性血管数 ;良性病变以规则型血管为主 ,CD34阳性血管数少于Ⅳ型胶原阳性血管数。结论 良、恶性病变的血管形态结构及数量有本质差别。CD34高表达是新生血管形成活跃的表现 ,说明肿瘤组织的高增殖性 ;Ⅳ型胶原高表达是血管发育成熟和结构完整的标志 ,说明肿瘤及其血管增殖缓慢 ;对瘤组织中微血管数和形态进行分析 。
Objective To investigate the relationship between the morphology and quantity of microvessels and the differentiation and proliferation of tumor cells in osteogenic tumor. Methods Morphological analysis and count of vessels with positive signal of CD34 and collagen Ⅳ in immunohistological staining were done in osteogenic tumor, ossifying fibrous dysplasia and normal bone callus. Results The form of vessels mainly were expansive blood sinus and slim bud. The number of positive vessels with CD34 was more than ones with collagen Ⅳ in osteosarcoma. The difference became more significant with neoplasm grading, nuclear division and the increase of PCNA and p53 expressions. It was opposite to the situation in benign pathology, and it took regular vessel as the dorminant factor. The number of positive vessels with CD34 were smaller than ones with collagen Ⅳ in benign disease. The difference was significant. Conclusion Not only the number of vessels in malignant and benign tumor were different, but also were the morphology and construction different in essence. The higher expression of CD34 showed active angiogenesis, indicating higher proliferation of tumor tissue. The higher expression of collagen Ⅳ protein was the mark for maturity of blood vessels and complete construction. It was an explanation for slow proliferation of tumor and blood vessel. The results are very important to understand the biological behavior of tumors and to guide clinical treatment.
