摘要
目的 研究全身放射损伤对皮肤伤口细胞外基质 (extracellularmatrix ,ECM)成分的影响及康复新 (W11-a12 )的促愈作用。 方法 10 0只雄性健康昆明种小鼠被随机分为单纯创伤组(SW组 )和全身放射损伤合并创伤组 (CRW组 ) ,每组 5 0只。CRW组采用 6Gy全身照射复合皮肤创伤模型。观察伤后 1,3,5 ,7,10d时伤口组织中Ⅰ、Ⅲ型胶原 (CL -Ⅰ、CL -Ⅲ )、弹力纤维 (EF)、纤维黏连蛋白 (FN)含量的变化及W11-a12 对其的作用。 结果 合并全身放射损伤后 ,皮肤伤口内几种基质成分的含量呈不同程度降低。其特点是 :(1)对不同的ECM成分抑制程度不同 ,按含量降低由重到轻依次是FN、CL -Ⅰ、EF、CL -Ⅲ ;(2 )对ECM不同成分抑制的持续时间不同 ,按降低时间由长到短依次是CL -Ⅰ、CL -Ⅲ、EF、FN。使用W11-a12 后 ,单纯皮肤创伤和合并全身放射损伤的皮肤伤口中几种主要ECM成分的含量均显著增高。 结论 伤口内ECM成分含量减少是合并放射损伤后伤口难愈的主要原因之一 ,W11-a12
Objective To investigate the effects of 6 Gy whole body irradiation on several components of wound extracellular matrix (ECM) and the repair promoting action of W 11 a 12 . Methods After mice were received 6 Gy gamma ray irradiation, the content of collagen I (CL I), collagen Ⅲ(CL Ⅲ), elastic fiber(EF), and fibronectin(FN) in wound tissue were observed. Results The contents of various ECM components in wound tissue were all inhibited after 6 Gy irradiation. The depression of various ECM components was variable and the order of depressing extent from the most to the least was FN, CL I, EF, and CL Ⅲ. The reduction of content among various ECM component was variable and the order of inhibiting duration from the longest to the shortest was CL I, CL Ⅲ, EF, and FN. W 11 a 12 increased the content of various ECM components both in simple wound tissue and in radiation impaired wound tissue. Conclusions The decrease of wound ECM contents is one of the major factors for the healing deficits of radiation impaired skin wound and W 11 a 12 has a good promoting effect on synthesizing and secreting ECM both in simple wound and in radiation impaired wound tissues.
