摘要
目的 探讨 E-选择素 S12 8R多态性与冠心病的关系。 方法 用聚合酶链反应 -限制性片段长度多态性 (PCR- RFL P)方法探查 E-选择素 S12 8R多态性在对照组和冠心病组中的基因频率分布。 结果 冠心病组 R等位基因频率高于对照组 (11.96 % vs 4 .71% ,P<0 .0 5 ) ,SR+RR基因型者空腹血浆 E-选择素水平高于 SS基因型者 (33.2 3± 7.2 0 vs 2 2 .5 8± 8.2 8ng/ m l,P<0 .0 5 )。 结论 E-选择素 S12 8R多态性与冠心病的发生有关 。
Objective\ To investigate the association of E\|selectin S128R polymorphism with coronary artery disease(CAD).\ Methods\ Genomic DNA was extracted from peripheral blood leukocytes in 92 CAD patients and 85 control subjects.\ DNA was amplified by polymerase chain reaction(PCR).\ PCR product was digested with the restriction enzyme PstⅠ and separated on 2% agarose gel electrophoresis.\ Concentrations of plasma soluble E\|selectin were estimated using the ELISA kits in 36 CAD patients and 29 control subjects.\ Results\ The frequency of R allele in CAD patients was significantly higher than that in controls(11 96% vs 4 71%, P<0 05).\ E\|selectin levels was related to E\|selectin genotype, being higher in subjects possessing the R allele(33 23±7 20 vs 22 58±8 28 ng/ml, P<0 05).\ Conclusion\ E\|selectin S128R polymorphism is associated with CAD and R allele may be a risk factor for CAD, involved in the onset of CAD with other risk factors such as high triglyceride and systolic pressure.
