摘要
目的 探讨 Th1 / Th2细胞因子水平变化在皮炎湿疹发病机制中的作用 .方法 收集 48例皮炎、湿疹患者外周血 ,用 Ficoll- Hypaque分离单个核细胞 ,采用夹心 EL ISA方法检测单个核细胞培养上清中的细胞因子 IFN- γ,(γ-干扰素 ) ,IL - 4(白细胞介素 - 4) ,IL - 1 0 (白细胞介素 ) .结果 皮炎、湿疹组患者 IFN- 水平高于正常对照组 (P<0 .0 5 ) ,其浓度(μg· L- 1)分别为 6 7.7± 2 7.3和 6 5 .2± 2 9.9,对照组浓度(μg· L- 1)为 1 5 .9± 8.2 ;IL- 4水平低于正常对照组 (P<0 .0 5 ) ,皮炎湿疹组浓度 (μg· L- 1)分别为 3 5 .6± 8.9和41 .6± 1 1 .8,对照组浓度 (μg· L- 1)为 85 .2± 2 2 .1 ;IL- 1 0水平在两组间无显著差异 ,皮炎湿疹组浓度 (μg· L- 1)分别为76 3 .3± 1 42 .2和 789.9± 1 3 1 .6 ,对照组浓度 (μg· L- 1)为72 9.6± 1 6 8.5 .结论
AIM To study the role of IFN γ, IL 4 and IL 10 cytokines in the process of dermatitis and eczema. METHODS Peripheral blood of contact dermatitis and eczema patients was collected and PBMC were separated with Ficoll Hypaque, BA ELISA was applied to detect cytokines IFN γ, IL 4, IL 10 from supernatants of PBMC culture. RESULTS Cytokines IFN significantly increased in contact dermatitis and eczema patients compared with normal controls ( P <0.05), the concentration (μg·L -1 ) was 67.7±27.3 and 65.2± 29.9 in contact dermatitis and eczema patients, and 15.9±8.2 in normal controls; while the level of cytokine IL 4 was lower in patients ( P <0.05), the concentration (μg·L -1 ) was 35.6±8.9 and 41.6±11.8 in contact dermatitis and eczema patients, and 85.2±22.1 in normal controls. However, there was no difference in the IL 10 level between patients and normal controls, the concentration (μg·L -1 ) was 763.3±142.2 and 789.9±131.6 in contact dermatitis and eczema patients, and 729.6±168.5 in normal controls. CONCLUSION Cytokines of Th1 play an important role in the development of dermatitis and eczema.
