摘要
目的 了解银屑病患者外周血单个核细胞 (PBMCs)对自体表皮角质形成细胞 (KCs)增生的作用 .方法 分离 3例银屑病患者的 PBMCs,经 3 0 Gy钴照射后与来自同一患者的KCs进行混合培养 ,观察 KCs增生代谢方面的变化 .结果 经过混合培养 ,来自银屑病患者皮损和皮损周围未受累皮肤的 KCs在自体 PBMCs作用下均发生快速增生反应 .结论 表皮 KCs与浸润炎性细胞的相互作用可能诱发 KCs的过度增生 。
AIM To learn the effect of psoriatic peripheral blood mononuclear cells (PBMC) on the proliferation of autologous epidermal keratinocytes. METHODS Peripheral blood mononuclear cells were isolated from 3 patients with psoriasis vulgaris. After irradiating in Cobalt gamma ray of 30 Gy, the cells were cocultured with psoriatic epidermal keratinocytes that were obtained from the same patient. The changes of keratinocyte proliferation were detected by 3H TdR incorporation assay. RESULTS Keratinocytes involved and uninvolved in Psoriatic underwent a significant proliferation response to autologous peripheral blood mononuclear cells in the mixed cultures. CONCLUSION Interaction of keratinocytes with infiltrated mononuclear cells in epidermis may induce the hyperproliferation of the keratinocytes and thus play an important role in the pathogenesis of psoriasis.
