白念珠菌钙调蛋白定点突变对重组菌株形态与生长的影响

摘要 目的 :研究白念珠菌钙调蛋白 (Cm d1)定点突变对重组菌株生长状态的影响。 方法 :以不同的培养温度培养 Cmd1定点突变重组菌株 ,分析其温度敏感性 ,在光镜下观察 Cmd1定点突变后重组菌株形态发生和生长速度的变化。结果 :重组菌株 B(F92 +)和 E(F89+F141+)菌株具有温敏性 ,在适当温度下才能生长 ,而重组 A(F89+)和 C(F141+)菌株无温敏性。光镜观察发现对照组、重组菌株 A和 C细胞芽体呈圆形或椭圆形 ,芽颈较短 ;而 B和 E菌株可见多数为细长的芽体 ,但也可见少许为圆形和椭圆形的芽体。 B和 E菌株培养早期的菌体总数明显少于对照组、A和 C菌株的菌体总数 (P<0 .0 5 )。结论 :Cmd1F92 +和 F89+F141+定点突变导致重组菌株具有温度敏感性 。 Objective: To study the effects of Candida albican calmodulin fixed point mutant on growth of recombinant strains. Methods: We studied temperature sensitivity of the Cmd1 fixed pointed mutants by culturing them at different temperatures, and observed the mutant morphogenesis and growth velocity with light microscope. Results:Recombinant mutant B and E strains were temperature sensitive yeast strains, which could grow only at appropriate culture temperature. Recombinant mutant A and C strains were not temperature sensitive strains. Under light microscope, control, A and C strain buds were spherical and oval, the bud necks were fairly shorter; meanwhile, some buds of B and E strian were slender, some of those were spherical and oval. In the earlier period of culture, the B or E strain were fewer than those of control, A or C strain ( P <0.05). Conclusion: Candida albican calmodulin F92 + and F89 +F141 + fixed point mutant result in the mutant temperature sensitivity and hysteresis of the morphogenesis and growth velocity. [
