递增剂量脂多糖致伤对大鼠SIRS肺损伤的影响 被引量:21

摘要 目的 梯级剂量脂多糖 (LPS)致伤对大鼠全身炎症反应综合征 (SIRS) 肺损伤严重程度的影响。方法 经Wistar大鼠尾静脉注射梯级剂量LPS( 2、4、6、8mg/kg)致伤 ,观察呼吸频率、死亡率 ,检测PaO2 、观察外周血WBC计数及分类大白细胞构成比、观察肺W/D值以及病理学检查。结果 ①LPS致伤 ,可以导致大鼠呼吸困难 ,PaO2 降低 ,外周血WBC计数异常 (LPS小剂量使其升高 ,大剂量使其降低 ) ,外周血大白细胞构成比升高 ,肺组织W /D值升高 ,肺病理改变加重 ;②LPS≥ 6mg/kg致伤 ,上述变化非常显著。结论 ①LPS致伤 ,可以诱发大鼠发生SIRS 肺损伤 ;②LPS 6mg/kg是导致大鼠SIRS 肺损伤失控 。 Objective To evaluate the effects of increased dose of lipopolysacchairde(LPS) on systemic inflammatory response syndrome(SIRS) acute lung injury(ALI) in rats. Methods Wistar rats were injured with increased dose of LPS(2,4,6,8 mg/kg) to cause the SIRS ALI. Respirtory rate, mortality, PaO 2,white blood cell(WBC) count in peripherl blood,proportion of large white blood cells,wet /dry weight(W/D) of lung and pathological observation were observed. Results ①Respiretory distress,decreased PaO 2,WBC disorder,increased proportion of large white blood cells, increased W/D value and pathological depravation was induced by LPS in rats. ②The changes of the above indexes were significant when LPS≥6 mg/kg was given to rats. Conclusion ①The SIRS ALI may be induced by LPS in rats. ②LPS≥6 mg/kg may cause ARDS in rats which is similar to that of the excessive expression of SIRS.
