内皮素-1促进去甲肾上腺素诱导的心肌细胞凋亡 被引量:6

摘要 目的 探讨内皮素 (ET- 1 )在去甲肾上腺素 (NE)诱导的心肌细胞凋亡中的作用 .方法 采用无血清培养新生大鼠心肌细胞模型 ,以免疫组化鉴定心肌细胞 ;以 Hoechst332 58与 PI双重荧光染色观察 ET- 1与 NE单独或联合作用时心肌细胞凋亡率的变化 .结果  1实验培养的新生大鼠心肌细胞的纯度达 90 % ;2 NE呈剂量依赖性诱导心肌细胞凋亡 ;ET- 1对心肌细胞凋亡率无明显影响 ;ET- 1与 NE联合作用可显著增加心肌细胞的凋亡率 .结论 内皮素 - AIM To study effect of endothelin 1 on norepinephrine induced apoptosis in cardiac myocytes. METHODS Primary cardiac myocytes were cultured in serum free media. These cells were identified by immunohistochemical staining. Cultured neonatal rat cardiac myocytes were treated with different concentrations of norepinephrine (NE) and ET 1 for 48 h and the apoptotic neonatal rat cardiac myocyte nuclei were visualized with the fluorescentDNA binding dyes Hoechst 33258 and propidium iodide under fluorescent microscope. RESULTS The purity of cultured neonatal rat cardiac myocytes was 90%; NE induced the apoptosis of cardiac myocytes in a dose dependent manner and ET 1 did not induce the apoptosis in cardiac myocytes; The treatment of 10 nmol·L -1 of ET 1 significantly enhanced NE induced apoptosis in cardiac myocytes. CONCLUSION ET 1 accelerates NE induced apoptosis in cardiac myocytes.
