摘要
应用TUNEL标记、DNA凝胶电泳以及光镜技术探讨去甲斑蝥素诱导人T淋巴细胞白血病细胞株 (6T CEM)凋亡规律。实验表明 :0 62 5~ 5mg L剂量的去甲斑蝥素作用 6T CEM细胞 2 4h及 1 0mg L剂量的去甲斑蝥素作用 6T CEM细胞 1 8h ,台盼兰染色阳性率均在 8 5 %以下 ;1 0~ 80mg L去甲斑蝥素作用 6T CEM细胞 2 4h明显抑制 6T CEM细胞生长。TUNEL标记显示 :1 0~ 40mg L剂量的去甲斑蝥素作用 6T CEM细胞 2 4h及 1 0mg L剂量的去甲斑蝥素作用 6T CEM细胞超过 1 8h后即出现典型的细胞凋亡特征。一定剂量范围内的去甲斑蝥素可在直接杀伤作用较小的情况下明显诱导 6T CEM细胞凋亡 ,不会因细胞坏死而引起炎症反应 。
To explore the apoptosis of human acute T lymphocyte leukemia cell line 6T CEM induced by Norcantharidin with TUNEL labeling, DNA gelelectrophoresis and microscopy. It was proved that trypanblue staining positive rates of 6T CEM cells exposed to Norcantharidin at the dosage of 0 625~5mg/L for 24 hours and 10mg/L for 18hours were all less than 8 5%;when treated by various dosage of Norcantharidin within range of 10~80 mg/L for 24 hours, the proliferation of 6T CEM cell was obviously inhibited. TUNEL labeling demonstrated that 6T CEM cells being treated by varied dosage of Norcantharidin for 24 hours showed typical features of cell apoptosis when dosage reached to 10mg/L;with dosage of Norcantharidin of 10mg/L, typical features of cell apoptosis were observed after 18 hours of treatment Dosage of Norcantharidin with minor killing efficacy had markedly induced apoptosis of 6T CEM cells without inflammation caused by cell necrosis. The result suggested a great potential of clinical application of Norcantharidin at a low dosage.
