摘要
棉酚已被认为是一种很有希望的男用抗生育剂,但它诱发的低血钾导致其不能进一步发展成为一种男用口服避孕药。棉酚诱发低血钾的机制虽已作了大量研究,但未被阐明。本文探讨并证明了棉酚及一些其它化合物对Ⅱ型11β-羟基甾体脱氢酶(11β-OHSD2)的抑制作用,并提出有关棉酚诱发低血钾机制的假说:棉酚抑制11β-OHSD2,引起肾脏中糖皮质激素的过量分泌,导致肾性失钾继而诱发低血钾。
Gossypol has been considered as a promising agent to induce infertility in many laboratory animals and in human kind, but the hypokalemia attributed to gossypol has inhibited further development of gossypot for use as a male oral contraceptive. The studies on the mechanism of hypokalemia induced by gossypol have not yielded a satisfac-tory explanation. We studied and proved the inhibition of type Ⅱ 11β-hydroxysteroid dehydrogenase (11 β-OHSD2)by gossypol and some other compounds,and our hypothesis remains that gossypol inhibits 11β-OHSH2 and causes gluco-corticoid excess in kidney ,leading to potassium lose and subsequent hypokalemia.
