高效液相色谱法测定人血清中盐酸阿夫唑嗪浓度

Determination of alfuzosin in Human Serum by HPLC

目的 :采用高效液相色谱法测定人血清中盐酸阿夫唑嗪的浓度。方法 :分析柱 :Shim -packCLC -ODS(4 6mm×150mm ,5μm )流动相 :甲醇 -乙腈 -磷酸盐缓冲液 -三乙胺 (10∶30∶60∶0 05) ;流速 :1 0ml/min ;荧光激发波长 :334nm ,发射波长 :378nm ;血清中样品用乙酸乙酯提取。结果 :血清中盐酸阿夫唑嗪浓度在0 40～51 20μg/L范围内线性关系良好 ,回归方程为 :A=0 3909C +0 0605(n=8 ,r=0 9996)。高、中、低3个浓度盐酸阿夫唑嗪的日内RSD分别为6 71 %、4 02 %和6 11 % ,平均回收率为100 87 % ;日间RSD分别为10 83 %、6 78 %和13 64 % ,平均回收率为97 93 %。最低检测血药浓度 (LOD )为0 20μg/L。结论 :本法灵敏、准确 ,分析时间短 。

OBJECTIVE:Alfuzosin concentration in human serum was determined by RP-HPLC METHODS:The mobile phase was methanol-acetonitrile-phosphate buffer-triethylamine(10∶30∶60∶0 05) After being extracted with ethyl ace_tate,alfuzosin was analyzed by reversed-phase HPLC (Shim-pack CLC-ODS,4 6mm×150mm,5μm)and fluorescence detection with excitation wavelength set at 334nm and emission at 378nm RESULTS:Over the concentration range of 0 4～51 2μg/L,the linear regression equation was A=0 3 909C+0 0 605(n=8,r=0 9 996) The average recovery of alfuzosin was 99 4% The intra-and inter-day RSDs were less than 15% CONCLUSION:The method is accurate and can be used for studying the pharmacokinetics of alfuzosin
