结肠癌及其癌前病变中bcl-2,bax蛋白的表达与细胞凋亡的关系

Study on relationship between expression of bcl-2,bax proteins and apoptosis in intestinal carcinoma and its precancerous lesions

[目的 ]研究结肠癌及其癌前病变中bcl 2 ,bax蛋白的表达与细胞凋亡的关系 .[方法 ]用免疫组织化学S P法检测bcl 2及bax蛋白 ,TUNEL法检测细胞凋亡 .[结果 ]细胞凋亡指数在腺瘤中表达最高 ,与正常组织中的比较有非常显著性差异 ,而在腺癌中表达最低 ,与腺瘤中的比较有非常显著性差异 ;bcl 2蛋白在腺癌中的表达率最高 (34 / 40 ,85 % ) ,其次是腺瘤 (10 / 15 ,6 6 % ) ,两者与正常及息肉组织比较均有显著性差异 ;bcl 2阴性的腺瘤凋亡指数与bcl 2阳性的腺瘤凋亡指数比较有显著性差异 ;bax蛋白阳性的腺瘤及腺癌凋亡指数与bax阴性的腺瘤及腺癌比较均有显著性差异 .[结论 ]bcl 2蛋白的过表达在结肠癌发生发展过程中起重要作用 ;bcl 2蛋白对细胞凋亡起抑制作用 。

OBJECTIVE To study the relationship between the expression of bcl-2, bax proteins and the apoptosis in intestinal carcinoma and its precancerous lesions. METHODS The bcl-2 and bax proteins were detected by immunohistochemistry and the apoptosis was detected by the TUNEL. RESULTS Apoptosis index was the highest in the adenoma and it was significantly different compared with the normal tissue, but the apoptosis index was the lowest in the adenocarcinoma and it was significantly different compared with the adenoma. The expression rate of bcl-2 protein was the highest in the adenocarcinoma(34/40,85%) and the next in the adenoma(10/15,66%), and the both rates were significantly higher than that in the normal and polyp tissue. Apoptosis index in the adenoma with negative expression of bcl-2 protein was significantly higher than that with positive expression of bcl-2 protein and in the adenoma and adenocarcinoma with positive expression of bax protein was significantly higher than that with negative expression of bax. CONCLUSION Over expression of bcl-2 protein plays an important role in the course of occurring and development of colon adenocarcinoma and the bcl-2 protein suppresses for apoptosis, but the bax protein accelerates for the apoptosis.
