对二氯苯对大鼠致畸作用的研究

摘要 [目的 ]研究对二氯苯 (PDCB)的致畸作用 ,为制定我国PDCB职业性接触限值标准提供参考依据。 [方法 ]妊娠的雌性SD大鼠随机分成 5组 ,每组 14~ 16只 ,实验组孕鼠分别以 3 60、180、3 6mg/kg剂量PDCB在妊娠的第 6~ 15d经口灌胃 ,阴性对照组孕鼠在妊娠的第 6~ 15d以 5ml/kg花生油灌胃 ,阳性对照组孕鼠在妊娠的第 9~ 11d以 7ml/kg敌枯双灌胃。在妊娠的第 2 0d处死孕鼠 ,分析胚胎发育指标与胎仔发育指标 ,检查有无外观畸形和骨骼畸形。 [结果 ]与阴性对照组比较 ,PDCB各实验组孕鼠体重增长未见显著减慢 ,活产仔数未见显著减少 ,死胎率与吸收胎率未见显著增加 ,胎仔平均体重、体长、尾长发育指标未见显著减缓 ,外观畸形与骨骼畸形未见显著增加。 [结论 ]在本实验剂量与条件下PDCB无母体毒性、胚胎毒性。 To study teratogenic effects of para dichlorobenzene (PDCB) in rats and to provide information for establishing occupational permissible exposure limit value of PDCB. Gravid female SD rats were divided randomly into five groups,14~16 rats per group. The rats in the experimental groups were administered with PDCB by gavage at a dos of 360,180,36 mg/kg per day respectively on day 6 through 15 of gestation. The rats in the negative control group were administered with peanut oil by gavage at a dos of 5 ml/kg per day on day of 6 to 15 of gestation. The rats in the positive control group were administered with N,N' methylene bis (2 amino 1,3,4 thiadiazole) at a dos of 7 mg/kg by gavage per day on day 9 to 11 of gestation. All animals were sacrificed on the 20 th day of gestation,embryonic developmental parameters and fetal developmental parameters were analysed,external and skeletal anomalies were examined. No significant differences were observed between the PDCB administered groups and the negative control group in body weight gain in pregnant rats,reduction of number of survivals at birth,increment of incidence of stillbirth,decrement of mean body weight,body length and tail length in the fetuses,retardation of skeletal development in the fetuses,increment of incidence of external and skeletal anomalies in the fetuses. [Conclusion] PDCB has no maternal toxicity,embryonic and developmental toxicity and teratogenicity under the experimental conditions.
