摘要 目的 :观察人牙乳头细胞内Smad1mRNA的表达及在BMP2作用下 ,细胞内Smad1mRNA的表达变化 ,探讨人牙乳头细胞内Smad1信号途径在BMP2调控牙乳头细胞分化中的作用。方法 :原代培养的人牙乳头细胞用BMP2处理后 ,提取总RNA ,采用Northernblot法 ,从mRNA水平观察Smad1基因的表达及含量变化。结果 :从mRNA水平观察到Smad1基因在人牙乳头细胞内的表达 ,但在BMP2作用 6、12、2 4h后 ,Smad1mRNA表达量无显著变化。结论 :人牙乳头细胞内存在Smad1信号转导途径 ,牙乳头细胞内Smad1mRNA表达量不受BMP2调控。 AIM: To investigate the expression of Smad1 gene and modulation in the human dental papilla cells (HDPC) treated by BMP2, and explore the molecular mechanism of Smad1 signaling during the differentiation of dental papilla cells into odontoblast regulated by BMP2. METHODS:Smad1 expression and modulation in the primary cultured HDPC treated by BMP2 was evaluated by Northern blot analyses.RESULTS: Cultured HDPCS express Smad1 protein, but the expression level of Smad1 was not significantly altered at 6h, 12h and 24h by BMP2 treatment. CONCLUSION: Smad1 signaling is present in the HDPC, but the expression level of Smad1 mRNA was not changed by BMP2.
