祁白术治疗脾虚证小鼠对消化器官组化和超微结构的影响

Changes of Histochemistry and Ultrastracture of Digestive Organ in the Spleen Difficiency Syndrome Mice after Treatment by Taking Medicine Qi Baizhu

目的 :探讨祁白术等治疗饮食失节所致脾虚小鼠模型的研究。方法 :模型小鼠随机分脾虚、祁白术、四君子汤、生及炒白术、正常、自然恢复等 7组。治疗前后分别取胰、肝、胃肠等进行组化、透射和扫描电镜观察。结果 :脾虚组胰腺泡上皮酶原、胰岛细胞、胃主及壁细胞、G细胞、内分泌细胞、十二指肠绒毛及吸收上皮微绒毛、小肠粘膜圆顶区 M细胞等均减少。固有膜呈慢性炎症 ,微循环瘀血明显。胰腺泡腔、上皮及胞核内有大肠杆菌感染。十二指肠绒毛及肠腺、肝及胰 ATP酶、SDH酶缺乏 ,肝苏丹 染色为脂肪变 ,肝、肌 PAS糖原染色减低、消失。经祁白术等治疗后上述病变皆有恢复。结论 :祁白术、四君子汤等对脾虚小鼠的糖、脂肪、蛋白质代谢及肠道、全身粘膜系统免疫障碍有复健作用 ,特别是祁白术能使血脂、脂肪肝、血糖降至正常。

Objective:To investigate the changes of mice model of the spleen defficiency syndrome (SDS) caused by irregular eating and treated by using Qi Baizhu (QBZ) and Si Junzi Tang(SJZT)etc.Methods:The mice were divided into 7 groups such as SDS,QBZ,SJZT,raw and parched Bai zhu, normality,natural cure at random, then comparison with all the groups each other were observed and studied on histochemistry,transmissional and scanning electron microscopy of the pancreas,livers,stomaches,intestines before and after the treatment by using QBZ and SJZT etc.Results:In SDS group,the zymogen granules in the epithelium of pancreas acini,pancreatic islet cells,the chief cells and parietal cells of stomaches, G cells, the other endocrine cells,the villi of duodenums, the microvilli of absorbing cells and the M cells at the round top in mucosas of small intestines were all decreased.The lamina propria had chronical inflammation and its microcirculation had stasis of blood vessels obviously,also there were infection with colon bacilli in the epthelium and its nucleus and acini lumen of pancreas .The enzyma of ATP,SDH were diminished in the villi of duodenuns, intestinal glands,livers and pancreas.The staining method sudanⅢ in livers were showed as fat degeneration,staining PAS of glycogen in livers and muscles were decreased significantly or disappeared .After the treatment of QBZ and SJZT etc.,above all the pathological changes were recovered and the M cells were increased. Conclusion: The Chinese herb QBZ and SJZT are an effective drug for treatment of SDS.It not only strengthens the metabolism of carbohydrate,fat and protein etc.but also increases the immumity of intestine and the mucosas system of all body. The QBZ can specially treat high blood fat,liver fatty and carbohydrate metabolism to be normal.
