端粒酶催化亚单位基因hTERT反义寡核苷酸与顺铂对Raji细胞协同作用的研究

摘要 目的 :研究端粒酶催化亚单位基因hTERT反义寡核苷酸可否增强Raji细胞对顺铂 (CDDP)的敏感性。方法 :hTERT基因的反义寡核苷酸作用Raji细胞 2 4h后 ,加入CDDP ,用台盼蓝拒染的方法计数活细胞、计算对细胞生存的抑制率。结果 :单纯hTERT反义寡核苷酸在浓度为 10 μmol/L时 ,对Raji细胞生存无明显抑制作用 ;但反义寡核苷酸加CDDP组对Raji细胞生存的抑制率明显高于随机序列 +CDDP组 (P <0 .0 5 ) ,其抑制作用在反义寡核苷酸作用 48h、CDDP作用 2 4h时最为显著 (P <0 .0 1)。结论 :hTERT基因的反义寡核苷酸与CDDP具有协同作用 。 Objective:To research whether the antisense oligonucleotides of telomerase catalytic sub-unit gene hTERT (human telomerase reverse transcriptase) can increase the susceptibility of Raji cells to cisplatin. Method:Raji cells were treated with cisplatin after exposed to hTERT antisense for 24 hours. The cell survival was determined by trypan blue dye exclusion test, and suppressive rate of cell survival was calculated. Result:Suppressive rate of cell survival in the presence of hTERT antisense and cisplatin was significantly (P< 0.05 ) higher than that of control cells which were treated with random oligonucleotides and cisplatin, and the most dramatic effects were seen at 48 hours using the antisense (P< 0.01 ). Conclusion:The hTERT antisense appears to have synergy with cisplatin, and it can increase the susceptibility of Raji cells to cisplatin.
