Expression of Fas/FasL antigen on peripheral blood T lymphocytes from uveitis patients

目的 :评价和探讨Fas/FasL抗原在葡萄膜炎患者外周血T淋巴细胞上的表达及其可能存在的意义。方法 :采用流式细胞术 (flowcytometry,FCM)及三色荧光双标记细胞免疫荧光染色技术对 2 6例Behc,et病、17例Vogt 小柳原田综合征患者、2 5例特发性前葡萄膜炎患者及 43例正常人外周血CD4 + T细胞和CD8+ T细胞表面Fas和FasL的表达进行检测。结果 :Behc,et病、Vogt 小柳原田综合征和特发性前葡萄膜炎患者CD4 + T细胞Fas的表达阳性率分别为 (2 5 .70± 7.32 ) %、(19.6 0± 11.0 2 ) %、(2 0 .81± 7.40 ) % ,均显著高于正常对照组[(14.0 2± 6 .30 ) % ],而FasL的表达阳性率与正常对照组比较 ,差异无显著性意义。Behc,et病和Vogt 小柳原田综合征患者CD8+ T细胞Fas的表达阳性率分别为 (9.47± 6 .97) % ,(6 .84± 5 .5 3) % ,均显著高于正常对照组[(3 .47± 2 .75 ) % ];特发性前葡萄膜炎患者CD8+ T细胞Fas的表达阳性率 [(5 .34± 3.0 8) % ]与正常对照组[(3.47± 2 .75 ) % ]比较 ,差异无显著性意义。Behc,et病和Vogt 小柳原田综合征患者CD8+ T细胞FasL的表达阳性率分别为 (3 .6 6± 2 .16 ) %、(4.0 5± 1.38) % ,与正常对照组 [(3.0 9± 2 .0 4) % ]比较 ,差异无显著性意义 ;特发性前葡萄膜炎患者CD8+

Objective:To evaluate the expression of Fas/FasL antigen on peripheral blood T lymphocytes and its possible implications for uveitis patients.Methods:The expression of Fas and FasL on peripheral blood T lymphocytes was determined using flow cytometry(FCW) and three color double marked immunofluorescence methods in 26 patients with Beh c[DD(;-1.6mm] ,et's disease(BD), 17 patients with Vogt Koyanagi Harada(VKH) syndrome, 25 patients with idiopathic anterior uveitis and 43 healthy individuals.Results:The expression of Fas antigen on CD 4 + T lymphocytes from patients with BD,VKH syndrome and idiopathic anterior uveitis(25.70±7.32)%,(19.60±11.02)%,(20.81±7.40)%,respectively was significantly higher than that in control group(14.02±6.30)%,but no difference was found in the expression of FasL antigen between patients and controls. The expression of Fas antigen on CD 8 + T lymphocytes from patients with BD or with VKH syndrome(9.46±6.97)%,(6.84±5.53)% respectively was significantly higher than that in control group(3.47±2.75)%. No difference was found in the expression of Fas antigen between patients with idiopathic anterior uveitis(5.34±3.08)% and the control group(3.47±2.75)%. There was no difference in FasL antigen expression for patients with BD or with VKH syndrome and the controls. However,lower expression of FasL on peripheral blood lymphocytes was noted in patients with idiopathic anterior uveitis(1.58±0.93)%.Conclusion:The expression of Fas antigen on CD 4 + T lymphocytes and CD 8 + T lymphocytes in uveitis patients was higher than that in controls. However,the expression of FasL antigen on these lymphocytes in patients was relatively insufficient. Presumably,the imbalance of the expression of Fas and FasL on T lymphocytes isresponsible for the perpetuation and recurrence of uveitis because the activated lymphocytes are not eliminated through apoptosis mediated by the Fas/FasL system.
