摘要
切取生长健壮的库拉索芦荟茎尖 ,在无菌条件下 ,接种至含有不同植物生长调节剂种类和浓度 ,在不同温度的 MS培养基中 ,进行诱芽和诱根培养 .以 MS+BA2~ 4+IBA0 .2~ 0 .2 5和 MS+BA2~ 6 +NAA0 .2~ 0 .3适合诱芽生长和继代培养 ;诱芽率 77.7%~ 1 0 0 % ;芽增殖系数 5 .66~ 6.0 0和 5 .44~ 6.3 3 ,以 MS+IBA2~ 4+NAA0 .2~ 0 .4适合诱根生长 ;诱根率达 3 0 6.6%~ 3 90 .0 % ,诱芽生长最适温度 2 5~ 3 0℃ ,诱根生长最适温度 2 0~ 2 5℃ ;完整组培苗移栽成活率达 92 % .
The stem tip culture of the adventitious buds and adventitious roots for Aloe vera L. has been made in MS culture media under the conditions of different sorts and concentrations of plant growth regulator and different temperatures. It shows that MS+BA 2~4 +IBA 0.2~0.25 and MS+BA 2~6 +NAA 0.2~0.3 are the best suitable regulators for adventitious budand for subclture buds respectively, with the inducing rate of bud of 77.7%~100% and the multiplication coefficient of 5.66~6.00 and 5.44~ 6.33 , and that MS+IBA 2~4 +NAA 0.2~0.4 is suitable for adventitious roots with the inducing rate of roots of 306.6% ~390.0%. Temperatures 25~30 ℃ and 20~25 ℃ are best for adventitious buds and adventitious roots respectively. Finally the surviving rate of transplant reaches 92%.
