摘要
用薄层色谱法 (TLC) ,以 ψ(石油醚∶乙酸乙酯∶苯 ) =70∶5∶1 5为展开剂 ,质量分数为 5 %香草醛硫酸为显色剂对木香超临界萃取物中的木香烃内酯与去氢木香内酯进行鉴别 ;并建立高效液相色谱法同时测定木香烃内酯与去氢木香内酯的含量方法。色谱柱为MerckLichrocartC1 8;流动相为 ψ(甲醇∶水 ) =70∶30 ;检测波长为 2 2 5nm。测得木香烃内酯和去氢木香内酯的线性范围分别为 0 41 78~ 2 .0 888μg和 0 .3950~ 1 .9752 μg ;回收率分别为 1 0 0 .3 %与 99.4 %。方法简便、灵敏。
The costuslactone and dehydrocostuslactone in the supercritical CO 2 extract of aucklandia were identified by TLC using a mobile phase of petroleum ether,ethyl acetate and benzene (70∶5∶15) and the colour developing reagent of 5% vanillic aldehyde dissovled in sulfuric acid. The contents of costuslactone and dehydrocostuslactone were determined by HPLC on a Merk Lichrocart C 18 column using a mobile phase of methyl alcohol and water (70∶30) with the 225nm detecting wavelength . The linear concentrations of costuslactone and dehydrocostuslactone were 0.4178 ~ 2.0888 μg and 0.3950~1.9752μg respectively. Their recoveries were 100 3% and 99.4% respectively. This determination method was simple,sensitive and fast.
