生长抑素类似物诱导人胆管癌SK-ChA-1细胞调亡及调亡相关基因表达的研究

摘要 目的 探讨生长抑素类似物SMS2 0 1 995 (SMS)抑制人胆管癌生长时对SK ChA 1细胞调亡调控基因bcl 2和bax的作用。方法 用DNA凝胶电泳和流式细胞仪检测AnnexinV标记凋亡细胞技术研究SMS诱导细胞凋亡的情况 ;用免疫组化和原位杂交分析SMS凋亡调控基因bcl 2和bax的影响。结果 SMS处理SK ChA 1细胞后 ,AnnexinV标记的凋亡细胞增多 ,DNA凝胶电泳显示典型的凋亡梯状图谱 ,同时SMS可使细胞bcl 2蛋白表达下降 ,使bax蛋白和mRNA表达升高。结论 SMS能诱导SK ChA 1细胞发生凋亡 ,bax和bcl 2凋亡基因介导了SMS对SK ChA Objective To investigate the role of somatostatin analogue (SMS201 995, SMS) on apoptosis of human cholangiocarcinoma SK ChA 1 cells and on apoptosis regulated gene bcl 2 and bax since the analogue inducing the inhibition of human cholangiocarcinoma growth. Methods DNA gel electrophoresis, flow cytometry with Annexin V as marker were used to detect the apoptosis of SK ChA 1 cells. The expressions of bcl 2 and bax were studied with SABC immunohistochemistry and in situ hybridization. Results The annexin V positive cells were increased after SMS treatment and showed a typical ladder pattern as apoptosis feature on gel. Besides, SMS inhibited bcl 2 protein expression and enhanced bax protein and mRNA expressions. Conclusion SMS can induce SK ChA 1 cell apoptosis by means of affecting bcl 2 and bax gene expressions.
