一个新的人乳源性生物活性肽cDNA克隆与测序分析

THE CLONING AND SEQUENCING OF A PROLACTIN -LIKE cDNA IN HUMAN MILK

目的:分离、克隆人乳汁中新的生物活性肽。方法:从人初乳中提取总RNA,反转录获得cDNA,SMARTPCR扩增cDNA片段,克隆于pGEM-TEasy载体中,进行测序分析。结果:克隆一个新的cDNA并编码42个氨基酸的多肽,与人催乳素样糖蛋白部分同源,加入国际基因库,编号为:GenbankAF160979。结论:人初乳中存在核酸RNA,所克隆的新cDNA可能具有催乳素样功能。

Objective:To isolate and clone a new cDNA in human milk.Method:The milk RNA was pu rified and re versed to cDNA.SMART PCR was used to construct the Library.Result:By DNA se quencing,a new cDNA en coding a predicted peptide of42residues was revealed,which shared41.5%amino acid and identified with prolactin-Like pro tein.Conclu sion:These data suggest that the predicted pep tide may play important roles in reproductive physiology.
